چکیده

During the last two decades, over 800 phytoplasma strains have been reported in association with several hundred plant diseases and numerous insect vectors. Research has yielded new knowledge about phytoplasma ecology and phylogenetic relationships. A taxonomic system has emerged, and perspectives on phytoplasma speciation have changed. Guidelines for naming new ‘Candidatus Phytoplasma’ species have been proposed; the16S rRNA gene was employed as the sole phylogenetic marker for species delineation. To date, 26 ‘Ca. Phytoplasma’ species have been named, forming the core framework of the emerging phytoplasma taxonomy. A classification system based on restriction fragment length polymorphism (RFLP) analysis of 16S rRNA gene sequences provides a simple, reliable, and rapid means to classify phytoplasmas on a large scale without a need to sequence the gene. The capacity of this classification system has been recently upgraded by the development of a computer-simulated RFLP analysis method. This approach has led to construction of the most comprehensive classification system for phytoplasmas to date. The concept of multi-gene sequence analysis for distinguishing phytoplasma species has emerged with the aim of overcoming deficiencies of the highly conserved 16S rRNA gene for delineating closely related phytoplasma species. Less conserved genes, such as ribosomal protein or secY, serve as phylogenetic markers for finer distinctions among phytoplasmas.

Phytoplasmas–The “Crouching Tiger” Threat of Australian Plant Pathology

Phytoplasmas are insect-vectored bacteria that cause disease in a wide range of plant species. The increasing availability of molecular DNA analyses, expertise and additional methods in recent years has led to a proliferation of discoveries of phytoplasma-plant host associations and in the numbers of taxonomic groupings for phytoplasmas. The widespread use of common names based on the diseases ...

Multiplex Real-Time qPCR Assay for Simultaneous and Sensitive Detection of Phytoplasmas in Sesame Plants and Insect Vectors

Phyllody, a destructive and economically important disease worldwide caused by phytoplasma infections, is characterized by the abnormal development of floral structures into stunted leafy parts and contributes to serious losses in crop plants, including sesame (Sesamum indicum L.). Accurate identification, differentiation, and quantification of phyllody-causing phytoplasmas are essential for ef...

Phytoplasma classification: Taxonomy based on 16S ribosomal gene, is it enough?

The discovery in 1967, of a new group of plant pathogens related to bacteria led to the finding of pleomorphic, wall-less prokaryotes in the phloem of many plant species affected by yellows-type diseases. Following the application of molecular technologies the phylogeny of these prokaryotes was resolved and led to the new trivial name of “phytoplasma” and to the designation of a new taxon named...
